Em 1766, o capitão-general de São Paulo, D. Luís Antônio de Souza Botelho Mourão, encarregou Antônio Corrêa Barbosa de fundar uma povoação na foz do rio Piracicaba. No entanto, o capitão povoador optou pelo local onde já se haviam fixados alguns posseiros e onde habitavam os índios Paiaguás, à margem direita do salto, a 90 quilômetros da foz, no lugar mais apropriado da região. A povoação seria ponto de apoio às embarcações que desciam o rio Tiête e daria retaguarda ao abastecimento do forte de Iguatemi, fronteiriço do território do Paraguai.
Oficialmente, o povoado de Piracicaba, termo da Vila de Itu, foi fundado em 1o. de agosto de 1767, sob a invocação de Nossa Senhora dos Prazeres. Em 1774, a povoação constitui-se freguesia, com uma população estimada em 230 habitantes, desvinculando-se de Itu em 21 de junho.
Em 1784, Piracicaba foi transferida para a margem esquerda do rio, logo abaixo do salto, onde os terrenos melhores favoreciam sua expansão. A fertilidade da terra atraiu muitos fazendeiros, ocasionando a disputa de terras. Em 29 de novembro de 1821, Piracicaba foi elevada à categoria de vila, tomando o nome de Vila Nova da Constituição, em homenagem à promulgação da Constituição Portuguesa, ocorrida naquele ano.
A partir de 1836, houve um importante período de expansão. Não havia lote de terra desocupado e predominavam as pequenas propriedades. Além da cultura do café, os campos eram cobertos pelas plantações de arroz, feijão e milho, de algodão e fumo, mais pastagens para criação de gado. Piracicaba era um respeitado centro abastecedor.
Em 24 de abril de 1856, Vila Nova da Constituição foi elevada à categoria de cidade. Em 1877, por petição do então vereador Prudente de Moraes, mais tarde primeiro presidente civil do Brasil, o nome da cidade foi oficialmente mudado para Piracicaba, "o mais certo, o correto e como era conhecida popularmente".

In 1766, the captain-general of St. Paul, D. Luis Antonio Botelho Mourão de Souza, Antonio Correa Barbosa instructed to establish a settlement at the mouth of the Piracicaba River. However, the captain opted for the place where settlers had already set a few settlers and the Indians where they lived Paiaguás, the right edge of the heel, 90 kilometers from the mouth in the most appropriate place in the region. On April 24, 1856, Vila Nova da Constitution was elevated to city status. In 1877, by application of then-Councilman Prudente de Moraes, later the first civilian president of Brazil, the city's name was officially changed to Piracicaba, "the most true, right and as it was popularly known."
The anniversary is commemorated in Piracicaba August 1.
